REKOMBINANT TRASTUZUMAB ANTITANASINI KODLOVCHI PLAZMID KONSTRUKSIYA YARATISH

Abstract
Saraton kasalligi tez o‘sib, 2020-yilda 18 million yangi holat va 10 million o‘lim qayd etilgan. 2040-yilga kelib, bu ko‘rsatkichlar 29,9 million va 15,3 millionga yetishi prognoz qilinmoqda. Ko‘krak bezi saratoni eng keng tarqalgan bo‘lib, 2022-yilda O‘zbekistonda 5 022 ta holat qayd etilgan. HER-2 (Inson epidermal o‘sish omili retseptori 2) geni amplifikatsiyasi ko‘krak bezi saratoni holatlarining 20-25% ida kuzatilib, o‘smaning tajovuzkor rivojlanishiga sabab bo‘ladi. Trastuzumab kabi dorilar samarali bo‘lsa-da, yuqori narx va talabning ortishi O‘zbekistonda uning mavjudligini cheklamoqda. Shu sababli, biz arzonroq mahalliy analog ishlab chiqishni maqsad qildik.
Buning uchun trastuzumab analoglari tahlil qilindi, va insonlashtirilgan trastuzumab oqsili genining nukleotid ketma-ketligi CHO hujayralarida ifodalanishiga mos ravishda kodon optimizatsiya qilindi. Virtual konstruksiya pVITRO1 plazmididan foydalangan holda ishlab chiqildi va yengil va og‘ir zanjir kodlash ketma-ketliklari uchun zaruriy elementlar kiritildi. Trastuzumabning og‘ir zanjir geni IgE signal peptidi bilan ishlab chiqilib, mEF1 promotor ostida, mEF-1a intronini o‘z ichiga olgan holda joylashtirildi, yengil zanjir esa rEF1 promotor ostida o‘sha IgE signal peptidi bilan kodlandi. Ushbu konstruksiya E. coli ga kiritildi, ijobiy klonlar PZR orqali genning kiritilgani tasdiqlandi.
